Hi,
In the first case, we're describing a location, so you need to use the dative case after "in", which is why you have the "en" ending on the possessive,
while in the second case, we're describing a direction, so you need the accusative case after "in", which is why you don't have the "en" ending (Büro uses neuter case so there's no mark in the accusative case).

Erratum: I just noticed I wrote "en" ending instead of "em" ending on "ihrem Büro".
