I think you could go either way in that example but I would put the "ne" before posso.
-Dove ne posso comprare. (Where can I buy it.)

I was thinking it could be "Dove posso comprarne una." But it does seem that we need "ne" somewhere??

I agree that "ne" is needed in this situation. My understanding is that while "Where can I buy one?" and "Where can I buy one of them?" are both perfectly acceptable in English, Italian requires the use of "ne", meaning "of them." My vote would be for peacemom's version: "Dove posso comprarne una?", but I believe "Dove ne posso comprare una?" is also correct.
