Because some words like "despues" require another word afterwords to validate what it is talking about. When using "despues" you should ask yourself "after what?" or "Despues DE que?" Well... "Despues de el museo" answers this question. When you have "de" and "el" together it forms a mandatory contraction... "del."

If you're saying that something is happening after something else, the phrase is "después de". So "after dinner" is "después de cenar." This phrase, then, would be "después de [after] el museo [the museum]". Then we condense "de el" down to "del" because that's the convention and it's a little easier to say that way.
